The realization of ABO blood grouping is based on two complementary tests:
- a globular test consisting of the search for antigens A (ABO1) and B (ABO2)
- a plasma test which consists of looking for anti-A and anti-B antibodies with A1 and B test cells
Anti-A and anti-B antibodies are regularly present in all individuals lacking the corresponding antigen.
The natural antibodies are spontaneously agglutinants in saline medium, their thermal optimum is at + 4 ° C, they have no hemolyzing power.
