Principle
The Eosin is an acidic dye that interacts with the cytoplasmic proteins rich in basic amino acids to form a complex red-pink brilliant.
The Eosin staining (cytoplasmic staining) in combination with the Haematoxylin (nuclear staining) is the most used method in the histopathological routine.
Method
1) Sections to water
2) Mayer’s Hemalum, 5 minutes
3) Tap water (or Scott’s water for 1 minute), 5 minutes
4) 1% eosin y aqueous solution, 5 minutes
5) Tap water, 5 minutes
6) Dehydrate
7) Clearing agent and mount.
Results
Nuclei: purple
Cytoplasm: pink-red
