Principle The Eosin is an acidic dye that interacts with the cytoplasmic proteins rich in basic amino acids to form a complex red-pink brilliant . The Eosin staining (cytoplasmic staining) in combination with the Hematoxylin (nuclear staining ) is the most used method in the histopathological routine. Method 1) Sections to water 2) Mayer’s Hemalum, 5 minutes 3) Tap water (or Scott’s water for 1 minute), 5 minutes 4) Washing in distilled water 5) Ethanol 80% 1 minute 6) (optional step to preserve the alcoholic content of the solution of Eosin-Phloxine) 7) Eosin-Phloxine alcoholic solution, 2-3 minutes 8) Ethanol 95 % x 2, 1 minute 9) Ethanol 95%, 1 minute 10) Absolute ethanol, 2 minutes 11) Absolute ethanol, 2 minutes 12) Clearing agent and mount.
