The animal-free Endosafe® Trillium™ rCR cartridges use an optimized kinetic chromogenic recombinant cascade reagent (rCR) developed to simulate the Limulus polyphemus LAL reaction. They provide an automated solution for quantitative bacterial endotoxin testing compatible with Endosafe cartridge platforms, delivering results in approximately 15 minutes.

Cartridge functionality

• Four reservoirs per cartridge: duplicate sample and spike channels for automatic duplicate runs as required by BET protocols
• Instrument draws and mixes sample with rCR reagent in sample channels
• Spike channels mix sample with rCR reagent and positive control for recovery checks
• Each sample is combined with chromogenic substrate and transferred to an optical cell for incubation and measurement
• Optical cells allow instrument measurement of color intensity and kinetic reaction time
• Measured reaction times are compared to an archived standard curve to calculate endotoxin concentration

Documentation and validation
Technical and validation documentation describe primary alternate method validation and beta study results demonstrating equivalency to licensed kinetic LAL assays. A validation support package includes protocols, a primary validation summary report and guidance for alternative method validation and interference screening.
