Product overview
The test kit consists of freeze-dried urea-arginine culture broth and susceptibility test boards. The broth contains peptone, yeast extract, serum, growth factors and nutrients, including urea and arginine. Alkaline products from urea/arginine metabolism raise broth pH and change color from yellow to red. The broth also contains antibacterial agents to inhibit bacterial and fungal contaminants. Test board wells are used for controls and for identification, enumeration and antibiotic susceptibility testing of Ureaplasma urealyticum (UU) and Mycoplasma hominis (MH).

Test principle
The kit uses freeze-dried urea-arginine broth and susceptibility test boards. Metabolism of urea and arginine produces alkaline compounds that change broth color from yellow to red, indicating growth. Selective antibacterial agents in the broth suppress bacteria and fungi. Test board wells provide control positions and specific wells for UU and MH identification/enumeration and for antibiotic susceptibility determination.

• Male: urethral swab/secretions, prostatic fluid, semen, or urine sediment (collect 10 ml urine, centrifuge 2000 rpm for 10 minutes)
• Female: cervical secretions or amniotic fluid collected appropriately
• Or: 30–50 μl freshly cultured positive broth

Results explanation

• Identification and enumeration: read and record Ureaplasma urealyticum (UU) results at 24 hours; read and record Mycoplasma hominis (MH) results at 48 hours.
• Antibiotic susceptibility: wells in line A contain lower antibiotic concentrations and line B higher concentrations; comparison of A and B for the same antibiotic determines susceptibility (kit materials include example layouts and interpretation guidance).
