How the Fiber Photometry System WorksGenetically encoded calcium indicators (GECIs) change fluorescence with intracellular calcium transients, which correlate with neuronal activity. A two-wavelength GCaMP approach (isosbestic reference + calcium-dependent excitation) is used to reduce motion and autofluorescence artifacts: an isosbestic excitation captures calcium‑independent signal (reference) while a calcium-dependent excitation measures activity-related fluorescence. The system combines signals across fibers to assess coordinated activity across brain regions and during social or behavioral tasks.
