Single cell RNA sequencing usually requires as an input a single cell suspension prepared from fresh samples. For single cell sequencing of frozen tissues, we offer a specialized product, the GEXSCOPE® Single Nucleus RNA Library Kit. The GEXSCOPE® Single Nucleus RNA Library Kit is a complete solution for transforming tissues into single nuclei and converting the mRNA contained in the single nuclei into NGS libraries. The GEXSCOPE® Single Nucleus RNA Library Kit can not only be used to analyze frozen tissue samples, but it can also be applied for single nucleus sequencing of the frozen cells isolated by FACS sorting. In addition, the GEXSCOPE® Single Nucleus RNA Library Kit can also be utilized for the single cell analysis of the cells with a large diameter or irregular shape such as neurons. Example of an Experimental Dataset Frozen hippocampus tissue from mouse brain was processed using the GEXSCOPE® Single Nucleus RNA Library Kit. After nucleus isolation, partitioning and barcoding, a single nucleus library was sequenced, and the obtained data were clustered. Based on the expression profiles of marker genes, major cell subtypes (excitatory neurons, inhibitory neurons, glial cells, neuroblasts and oligodendrocytes) were annotated using the SynEcoSys database.
