Design and principleThe Key is supplied with immobilized Low Level Control (LLC), anti-cPL antibody and High Level Control (HLC) in an aluminum foil pouch with desiccant. The matched Cartridge contains separated reagent compartments sealed with protective foil. The assay principle relies on sequential incubation and transfer of the Key across compartments to allow antigen–antibody binding, labeling and color development on the Key surface.
Brief procedureFor each sample use one Key and one Cartridge. Deposit canine serum into top compartment 1 of the Cartridge and mix. Remove the Key from its foil pouch and insert into compartment 1; incubation causes disappearance of pink control spots as the specific antibody–antigen–antibody reaction proceeds. Transfer the Key to subsequent top compartments at timed intervals so that enzyme conjugate labeling occurs in compartment 3 and final substrate development produces a purple‑blue signal on the Key in compartment 6.
Wash stepsCompartment 2 removes unbound cPL antigen and sample matrix. Compartments 4 and 5 remove unbound or excess enzyme conjugate. Compartment 7 removes excess chromogen produced in compartment 6. A valid test shows distinct purple‑blue signals on both HLC and LLC control spots.
