Design and Principle
One Solid Array Unit, one Solution Unit and one Substrate are used per sample. The Solid Array Unit contains a membrane array with location markers and allergen spots, packaged in an aluminum foil bag with desiccant. The Solution Unit holds reagents in separate compartments of a plastic cartridge. The sample is deposited into compartment 1 and absorbed by the Solid Array Unit; subsequent timed transfers move the array through wash and reagent compartments. Bound feline IgE antibodies on the protein array are labeled via a biotin-streptavidin enzyme reaction (anti-feline IgE-biotin in compartment 3; streptavidin-enzyme in compartment 5). Wash steps in intermediate compartments remove unbound materials. Finally, substrate is added to develop purple-blue spots where enzyme is bound; visible purple-blue location markers on the membrane confirm valid performance.
