ALK is an anaplastic lymphoma kinase. The ALK gene encodes a transmembrane tyrosine kinase receptor . ALK gene rearrangement is a driving mutation underlying the development of NSCLC. EML4 is the most common ALK fusion partner. ALK gene rearrangement has been identified in 5–6% of NSCLC case. In 2017, the NCCN clinical practice guidelines clearly stated that ALK gene testing is necessary before treatment for NSCLC. Crizotinib is is an oral anaplastic lymphoma kinase (ALK) inhibitor for the treatment of ALK positive non-small cell lung cancer, which was approved by the US FDA in 2011. Crizotinib has shown significant benefits in the management of ALK positive NSCLC compared to conventional chemotherapy. DETECTION SIGNIFICANCE Qualitative detection of EML4-ALK gene fusion mutation in RNA samples is performed to evaluate the degree of benefit from ALK tyrosine kinase inhibitors (TKIs). This helps to assist clinicians in more comprehensively selecting NSCLC patients who may benefit from crizotinib, and is applicable to NSCLC patients before entering targeted therapy, providing a scientific basis for individualized treatment of cancer patients.
