PrincipleThe assay is based on competitive inhibition immunochromatography: Nitrofuran (AHD) in the sample binds colloidal gold-labeled monoclonal antibody, reducing antibody binding to immobilized Nitrofuran (AHD)-BSA on the test line. The resulting color intensity at the T line relative to the C line provides a qualitative result against the detection limit.
Sample preparation- Solution preparation: prepare 0.1M K2HPO4 by diluting 1M K2HPO4 with deionized water at 1:9.
- Egg sample treatment:
- 1) Weigh 3 ± 0.05 g homogenized egg sample into a 50 ml centrifuge tube; add 6 ml deionized water, 750 µl 1M HCl and 300 µl derivatization reagent; shake 2 min; incubate at 60 ℃ water bath for 30 min.
- 2) Add 7.5 ml 0.1M K2HPO4, 600 µl 1M NaOH and 10 ml ethyl acetate; shake 1 min; centrifuge at 4000 r/min for 10 min.
- 3) Transfer 5 ml upper organic layer to a glass tube and evaporate to dryness at 56 ℃ under nitrogen or air.
- 4) Add 2 ml n-hexane and shake to dissolve residue completely; add 0.6 ml sample buffer, shake 30 s, allow phase separation; remove upper layer (if emulsion forms, centrifuge 4000 r/min for 5 min).
- 5) Withdraw 200 µl of the lower aqueous layer for testing (avoid aspirating the upper organic layer).
Operation procedures- 1) Read the manual before use and bring dipsticks and samples to room temperature.
- 2) Prepare required bottles, microwells and dipsticks from the kit; mark samples. Use prepared dipsticks within 1 hour and keep reagent bottles sealed to avoid moisture.
- 3) Add 200 µl of processed sample into a microwell; aspirate and dispense 5–10 times to mix until no visible solids remain.
- 4) Incubate 5 min at room temperature (20–25 ℃), then insert a dipstick with the MAX end fully immersed in the mixture.
- 5) Incubate 5–8 min at room temperature; read result at this time only (other read times are invalid).
Test result interpretation- Negative: T line is darker than or equal in color to the C line — no Nitrofuran (AHD) residue or below detection limit.
- Positive: T line is obviously lighter than the C line or T line is absent — Nitrofuran (AHD) residue at or above detection limit.
- Invalid: C line absent — test invalid (expired or incorrect operation); repeat with a new test.
Precautions- Use each test strip once at room temperature; do not use after expiry date.
- Avoid reuse of pipette tips to prevent cross-contamination.
- Do not touch the central white membrane of the test strip; avoid direct sunlight and strong airflow during testing.
- Wear appropriate personal protective equipment (gloves, lab coat) when handling reagents and samples.
SpecificityNo cross-reaction observed with Nitrofuran AMOZ, AOZ or SEM under the stated conditions.
