Intended use and principle
For staining acid-fast Mycobacteria and other acid-fast bacteria; follows Ziehl–Neelsen/WHO carbofuchsin recommendation
Acid-fast organisms with lipid-rich cell walls retain carbolfuchsin after decolorization with acid-alcohol and remain red; non–acid-fast cells take up the blue counterstain
Staining result
Acid-fast bacteria (e.g. Mycobacterium tuberculosis) appear red on a blue background; other bacteria and cells appear blue
M. tuberculosis: typically rod-shaped, slightly curved; width ~0.3–0.6 μm; length 0.5–8 μm (most 1.5–3.5 μm). Well-stained organisms may show darker particulates and are visible singly or in clusters under 100× oil immersion
