Bacterial spores are the resting state of bacteria. They can resist various physical and chemical factors and endure execrable conditions. Spores, which have strong refraction, can be observed under light microscope when properly stained. Baso Spore Stain is a very useful tool for bacteria differentiation that allows the identification of size, shape and location of bacterial spores.
Methods:
1. Prepare smear of bacteria with spores, air dry and fix on flame.
2. Add Carbolfuchsin on slides. Heat on small flame and keep dye steaming for about 5 minutes. After cooling, wash with water.
3. Decolorize with 95% ethanol for 2 minutes and wash with water.
4. Counterstain with Alkaline methylene blue for 30 seconds, followed by water wash. Air dry slides and then observe under oil immersion lens.
