Principle
The device is a competitive immunoassay. Drug conjugates are immobilized on the test region of the membrane. During testing, the urine specimen mobilizes antibodies conjugated to colored particles on the sample pad. As the mixture migrates by capillary action, antibodies bind either to immobilized drug conjugates or to free drug molecules in the specimen. If drug concentration is below the cutoff, antibody‑colored particle conjugates bind to immobilized conjugates forming a visible colored band at the test region (negative). If drug concentration exceeds the cutoff, free drug competes for antibody binding sites and prevents band formation at the test region (positive). A colored band at the control region confirms proper specimen volume and membrane function.
