Method For histology
1) Sections to ethanol 95°
2) Sudan III, 10 minutes
3) Differentiate rapidly in ethanol 95°
4) Wash in distilled water
5) Mount in acqueous mount
For fecal staining
1) Add few drops of Sudan III to a thin layer of fresh fecal sample on a glass slide. Neutral fats turn orange in large droplets.
2) In case of suspected malabsorption syndrome, the sample must first be acidified with two drops of acetic acid then gently heated with a small flame and then colored with Sudan III. Non-absorbed fats are strongly orange colored.
3) A malabsorption syndrome is suspected when the fecal sample is negative for the first stain (point 1) and positive for the stain described in point 2.
