Acid-fast stain (Kinyoun) for automated stainers is a ready-to-use staining solution intended for medical diagnosis and research. Optimized for use on automated staining platforms, it differentiates acid-fast organisms such as Mycobacterium spp. by producing a red stain on acid-fast bacteria with a contrasting counterstain of background elements.

Testing principle

Acid-fast bacteria possess lipid-rich cell walls that resist conventional staining. When stained with a phenolic dye like carbolfuchsin, the dye–lipid complex resists decolorization by acid-alcohol, allowing acid-fast organisms to retain the red color and be distinguished from non–acid-fast cells.

Stain results

• Methylene blue counterstain: acid-fast bacteria appear red on a light blue background; other bacteria and cells appear blue.
• Malachite green counterstain: acid-fast bacteria appear red on a light green background; other bacteria and cells appear green.
• Typical morphology (e.g., Mycobacterium tuberculosis): slender bacilli ~0.3–0.6 µm width and commonly 1.5–3.5 µm length; well-stained organisms may show metachromatic granules and are observed under 100× oil immersion.
