Testing principle

• Acid‑fast bacteria stained with Auramine O exhibit bright orange to yellow‑green fluorescence under a fluorescence microscope using a violet/blue excitation filter (approx. 350–490 nm). The fluorescence signal is detectable at low magnification to facilitate rapid screening.

Stain results

• Acid‑fast mycobacteria appear as bright orange‑yellow to green‑yellow, slightly curved rods on a dark background; non‑specific background shows pale yellow staining.
