- Rapid Romanowsky-derived staining solution developed by Professor Liu Zhenhui for use with automated stainers. Produces Wright-like results in approximately 1–2 minutes and is indicated for blood smears, bone marrow, gynecological discharge and exfoliative cytology.
Testing principle
- Staining relies on dye penetration and retention through physical adsorption and chemical affinity. Different cells and cellular components present varying affinities for the acidic dye (eosin) and the basic dye (methylene blue) in Liu's solution, producing differential coloration that aids morphological identification.
Stain results
- Blood smear and bone marrow smear: red blood cells appear light red or light green; cytoplasmic granules of white blood cells are clearly visible with characteristic colours for each cell type; nuclei stain purple-red with clearly visible chromatin.
- Gynecological discharge staining:
- Trichomonads: typically pear-shaped, round, oval or irregular; cytoplasm gray-blue or sky-blue, with a small eccentric nucleus; flagella may sometimes be visible.
- Candida (yeast): stained dark blue with visible budding spores and slender pseudohyphae.
- White blood cells: morphology similar to blood smears; quantity indicates degree of inflammation.
- Other findings: Leptothrix (long, thin, gray-blue), Gonococci (blue, kidney-shaped, sometimes intracellular; caution advised to distinguish from staphylococci), Gardnerella (blue, coccobacillary).
- Exfoliated cell staining: nuclei stain purple-red and nucleoli stain blue; cytoplasm shows varying shades of blue according to differentiation (basophilia).
