Testing principle:Acid‑fast bacteria possess a lipid‑rich cell envelope that resists conventional staining. After staining with an enhanced dye (carbolfuchsin), the dye–lipid complex resists decolorization by acid‑alcohol, allowing acid‑fast organisms to retain the red stain and be differentiated from non‑acid‑fast microorganisms. This kit follows the Ziehl‑Neelsen (carbolfuchsin) principle recommended by WHO and uses an improved basic fuchsin method rather than fluorescent techniques.
Stain results:- Methylene blue counterstain: acid‑fast bacteria appear red on a light‑blue background; other bacteria and cells appear blue.
- Malachite green counterstain: acid‑fast bacteria appear red on a light‑green background; other bacteria and cells appear green.
- Typical Mycobacterium tuberculosis morphology: bacilli, slightly curved, width ~0.3–0.6 µm, length typically 0.5–8 µm (most 1.5–3.5 µm); well‑stained organisms may show metachromatic granules and are observable under 100× oil immersion.
