Testing principle
Cellular proteins and sperm components have different isoelectric points and dissociation behaviors under the same acidity conditions. Acidophilic proteins expose positively charged amino groups that bind acidic dyes (eosin) and appear red. Basophilic proteins expose negatively charged carboxyl groups that bind basic dyes (methylene blue) and appear blue. Neutrophilic components bind both dyes and show purple-red coloration. This differential dye binding underlies the Diff-Quik staining method recommended by WHO for rapid cytological assessment.

Stain results

• Blood smear: red blood cells appear red; leukocyte cytoplasmic granules are distinctly colored according to cell type; nuclei stain purple-red with visible chromatin detail.
• Semen smear: post-acrosomal region stains purple-blue and acrosomal region light purple-red; midpiece and tail show light red staining; various cellular elements display distinct morphologies and staining patterns.
