Overview

Clear colourful acid-fast stain reagents formulated for use with automated stainers to enable unambiguous identification of acid-fast bacteria. Testing principle

Acid-fast bacteria (e.g., Mycobacterium species including Mycobacterium tuberculosis) possess lipid-rich, waxy cell envelopes that resist decolorization by acid-alcohol after staining. The method applies the enhanced carbol fuchsin, decolorizes non-acid-fast organisms with acid-alcohol, then applies a methylene blue counterstain. Acid-fast organisms retain the primary red stain while non–acid-fast cells take the counterstain, allowing visual differentiation under light microscopy.

Stain results

Acid-fast bacteria appear red against a light blue background; other bacteria and tissue elements appear blue. Typical Mycobacterium tuberculosis morphology: primarily rod-shaped (slightly curved), width ~0.3–0.6 µm, length ~0.5–8 µm (commonly 1.5–3.5 µm). Well-stained preparations may show strongly stained metachromatic granules and, under 100× oil-immersion, individual bacilli, clusters or branching arrangements can be observed.
