Acid Fast Stain (Phenol-free Auramine O staining method) formulated for use with automated stainers to detect acid-fast bacteria by fluorescence microscopy. The reagent set uses Auramine O fluorescent dye (phenol-free formulation) so acid-fast organisms emit bright orange-yellow or yellow-green fluorescence under a 350–490 nm violet-blue excitation, enabling rapid detection at low magnification.

Testing principle

• After staining with Auramine O, acid-fast bacteria are examined with a fluorescence microscope equipped with an ultraviolet light source. Under a 350–490 nm violet-blue filter, acid-fast bacteria emit bright orange-yellow or yellow-green fluorescence.

Stain results

• Acid-fast bacteria: bright orange-yellow or yellow-green, rod-shaped, slightly curved, on a black background.
• Background: generally black; some residual materials may appear light yellow.
