Modified Sperm Pap Stain (for automatic stainers) by Baso is an accelerated, machine-adapted Papanicolaou (Pap) staining solution formulated for automated staining instruments to deliver clear, multicolour staining of sperm and semen cells. The method is a simplified Pap protocol modified to match the operational cycles of automated stainers and builds on the WHO-recommended Pap technique.

Testing principle

• Sperm components and cellular proteins have different isoelectric points and charges under identical pH conditions, enabling selective binding to specific dyes. Nuclei (mainly acidic substances) are stained with the basic dye hematoxylin, while cytoplasmic components (mainly basic substances) preferentially bind acidic dyes. The procedure is a simplified Papanicolaou stain adapted for automatic stainers.

Stain results

• The acrosomal region of the sperm head appears light blue or colorless/transparent; the post-acrosomal region stains dark blue. The midpiece and tail show light red or light green staining. Different semen cells exhibit variable morphology and dye affinity depending on shape and composition.
