Antigen BackgroundAlpha-synuclein is a 140–amino-acid neuronal protein of the synuclein family. It shares sequence homology with beta-synuclein and is conserved across vertebrates. Lacking a signal peptide, alpha-synuclein is predominantly intracellular and contains repeating motifs that form alpha-helical regions implicated in membrane interaction and synaptic localization. Aggregated and truncated forms have been identified in Lewy bodies and are associated with neurodegenerative pathology.
Specificity and applicationClone KM51 specifically recognizes alpha-synuclein and does not cross-react with beta-synuclein. It has been used to detect full-length, truncated and aggregated alpha-synuclein in human brain material, including immunohistochemical staining of Lewy bodies. For optimal detection in tissue sections, pretreatment with 98–100% formic acid is recommended.

Recommended pretreatment and protocol notesUse formic acid pretreatment (98–100%) for paraffin-embedded tissue sections as indicated. Consult the IFU for recommended dilutions, incubation times and validated procedures. This antibody has been reported in both immunohistochemistry (IHC) of Lewy body dementia tissue and Western blot analyses of purified Lewy bodies.
