Application and principleReference method for demonstrating protein deposits in renal biopsy. Recommended fixative: Bouin.
- Principle: nuclear staining with ferric hematoxylin, cytoplasmic staining with Orange G and highly selective collagen staining with aniline blue mediated by phosphomolybdic acid.
- Orange G does not bind phosphomolybdic acid and stains the remaining structures.
- Acid fuchsin highlights protein deposits in bright red.
- Results: collagen fibrils — blue
- nuclei — black
- erythrocytes, cytoplasm — pink-orange
- elastic fibers — pale pink-yellow or colorless
- protein deposits — bright red
Method (overview)- Hydrate section and apply reagents A and B (5 drops each) for 10 minutes, rinse.
- Apply reagent C (10 drops) for 5 minutes, rinse; apply reagent D (10 drops) for 5 minutes, rinse.
