It is the method of choice in liver biopsy. Result The removal of glycogen can be detected, after PAS reaction, by comparing the sample section with an adjacent section of the same preparation that has not been treated with diastase. Product for the préparation of cyto-histological samples for optical microscopy. Digestion of histologie sections by means of diastase solution is advisable when the purpose is to remove glycogen in order to observe épithélial neutral mucins. This method is recommended for liver biopsy. PRINCIPLE The Diastase essentially hydrolyses starch and glycogen on the sections of tissue. Usually digestion of glycogen is not complété on tissue fixed because of particular glycosidic bond that they are formed during conventional fixation and it is possible to observe a granular glycogen precipitate to a pôle of the hépatocytes.
