Intended use
For enzymatic digestion on histological sections to remove glycogen and allow specific observation of neutral epithelial mucins. Preferred method for liver biopsy specimens in routine histopathology.

Principle
Diastase hydrolyzes starch and glycogen in tissue sections. Digestion on fixed tissue may be incomplete due to particular glycosidic bonds formed during conventional fixation, with granular precipitation of glycogen at a pole of hepatocytes.

Method

Bring the section to distilled water.
Bring the diastase solution to room temperature.
Incubate the slide at room temperature for 30 minutes.
Rinse the slide several times in distilled water.
Proceed with PAS staining as usual.
