Application and result
Combined histological staining method for differentiation of acidic mucins, neutral mucins and carbohydrates on tissue sections. Expected staining: PAS-positive substances magenta/red; acidic mucopolysaccharides blue–turquoise; some acidic mucins and cartilage purple to dark blue.

Method

1) Hydrate section in distilled water.
2) Apply 10 drops of reagent A to the section: leave for 30 minutes.
3) Drain the slide without rinsing and apply 15 drops of reagent B to the section: leave for 10 minutes.
4) Rinse 5 minutes in tap water and 2 minutes in distilled water.
5) Apply 10 drops of reagent C to the section: leave for 10 minutes.
6) Rinse in distilled water.
7) Apply 10 drops of reagent D to the section: leave for 20 minutes.
8) Rinse in distilled water.
9) Apply 10 drops of reagent E to the section: leave for 2 minutes.
10) Drain the slide without rinsing and apply 10 drops of reagent F to the section: leave for 3 minutes.
11) Rinse in distilled water.
12) Apply 10 drops of reagent G to the section: leave for 2 minutes.
13) Differentiate in running tap water for 5 minutes.
14) Dehydrate through ascending alcohols, clear in xylene and mount.
