To demonstrate copper in hepatic tissue sections.
PRINCIPLE
Serious liver diseases are associated to an increase of copper, which is normally present in hepatic tissue. In Wilson disease, the decrease of biliary excretion of copper results in a copper accumulation within liver cells. DMAB-rhodanine (5-pdimethylbenzylidene-rhoadanine) binds both to the free copper quota and copper associated protein.
METHOD
1) Bring section to distilled water.
2) Preparation of Rhodanine solution: In a 50 ml Coplin Jar, put 40 ml of distilled water and add 1 ml of reagent A , 1 ml of reagent B and 20 drops of reagent C. Stir briefly with a glass stick.
3) Put the slide in the obtained solution and incubate in an oven at 56° C for 3 hours or at 37°C overnight.
4) Rinse slide in three changes of distilled water.
5) Put on the section 10 drops of reagent D: leave to act 3 minutes
6) Blueing for 2 minutes in running tap water.
7) Dehydrate through ascending alcohols, clear in xylene and mount.
8) It is recommended to read the slide with immersion oil and 100X.
