Intended use

Combined staining method for demonstration of Helicobacter pylori (blue) and epithelial mucins (yellow) on paraffin-embedded gastric tissue sections for optical microscopy.

Principle
Prior oxidation with periodic acid renders gastric epithelial mucins, normally not stained by Alcian-group dyes, highly reactive to Alcian yellow, forming an insoluble and stable complex. Bacteria are subsequently visualized using toluidine blue.

Method

1) Hydrate the section in distilled water.
2) Apply 10 drops of reagent A to the section; incubate 10 minutes.
3) Rinse thoroughly in distilled water.
4) Apply 10 drops of reagent B; incubate 5 minutes.
5) Rinse in tap water for 2 minutes.
6) Apply 10 drops of reagent C to the slide; incubate 5 minutes.
7) Rinse thoroughly in distilled water.
8) Apply 8 drops of reagent D and 2 drops of reagent E to the section; incubate 3 minutes.
9) Rinse thoroughly in distilled water.
10) Air dry.
11) Dehydrate in alcohol, clear in xylene and mount.
