To demonstrate elastic fibres in tissue sections.
PRINCIPLE
The method is based on the affinity towards elastic fibres displayed by resorcin fuchsin, a precipitate resulting from a reaction between resorcin, basic fuchsin and ferric chloride. Since this is not a specific method, other structures such as collagen and basal membranes might be stained. Therefore, it is essential to differentiate carefully in order to obtain a selective marked staining of elastic fibres. Weigert’s “long method” differs from “quick method” in both staining mixture and procedure; it is more selective because staining mixture is less concentrated and incubation times are very long.
METHOD
1) Bring section to distilled water.
2) Put on the section 5 drops of reagent A and add 5 drops of reagent B: leave to act for 5 minutes.
3) Rinse in distilled water.
4) Put on the section 10 drops of reagent C, leave to act for 5 minutes.
5) Rinse in distilled water.
6) Transfer the reagent D in a Coplin jar, introduce the slide, cap the jar and incubate overnight. After use, in order to reduce the ethanol evaporation, it is recommended to re-introduce the solution in the original vial.
7) Rinse in distilled water. Put on the section 10 drops of reagent E: leave to act 10 minutes.
8) Rinse well in distilled water.
9) Dehydrate through ascending alcohols, clear in xylene and mount.
