Description

Product for the preparation of cyto-histological samples for optical microscopy. Application Cryostatic sections of 8 microns of human skeletal muscle. Histochemical demonstration of esterase in denervated fibres. Method 1) It is recommended to let the reagents reach room temperature before starting the procedure. 2) The reagent D contains isopropanol. To avoid the complete evaporation of this solvent it is recommended not to open the vial until the moment of utilization (step 3 of the method). 3) Preparation of incubating solution: - transfer reagent D into the tube A. Shake 5-10 seconds. - add reagent B to reagent C. Shake 5-10 seconds and allow to stand for 1 minute. - put B+C solution into the tube A (A+D solution) and shake shortly. 4) Incubate sections at 37°C, 60 minutes. 5) Rinse in distilled water, 3 changes 6) Dehydrate starting from ethanol 90% clear and mount. Results Angular atrophic fibres: beige Neuromuscular junction: brown Lipofuscin: brown Lysosomal activity: brown Warning and precaution The product is intended for professional laboratory use for healthcare professionals.
