Description

Product for the préparation of cyto-histological samples for optical microscopy. To demonstrate elastic fibres in tissue sections. Recommended for vascular pathology. PRINCIPLE The method is based on the affinity towards elastic fibres of the hematoxylin with Lugol solution. This is not a spécifie method, other structures such as collagen and muscle tissue might be stained. Therefore, it is essential to differentiate carefully, in order to obtain a sélective marked staining of elastic fibres. Counterstaining with Sirius red picrate solution makes it possible to differentiate the connective tissue. METHOD 1) Bring section to distilled water. 2) Put on the section 10 drops of reagent A: leave them act 30 minutes. 3) Rinse in distilled water. 4) Tap water 3 minutes. 5) Introduce the slide in the incubation box and put on the section 8 drops of reagent B + 4 drops of reagent C + 4 drops of reagent D. Leave to act at room temperature for 25 minutes. 6) Rinse in distilled water. 7) Differentiate with reagent E: 2 or 3 changes of 15 seconds each. 8) Rinse carefully in distilled water. 9) Put on the section 10 drops of reagent F: leave to act 1 minute. 10) Rinse in distilled water. 11) Dehydrate through ascending alcohols, clear in xylene and mount Product classification The product is intended for professional laboratory use for healthcare professionals.
