Description

Product overview
Staining kit for detection of AgNOR (argyrophilic nucleolar organizer region) proteins (100 kD) in paraffin-embedded sections and cytology smears. Reference: 04-045801. Supplied for 48 tests. Minimum number of preparations: 12 (up to 4 slides per preparation). Total processing time per run: 30 minutes.

Principle
AgNOR proteins (100 kD) act as reducing agents that bind silver ions and produce a metallic silver deposit visible under optical microscopy. Immediately before use, silver nitrate solution is combined with a positively charged hydrocolloid (e.g., gelatin) to enhance contact between reducing groups of nucleolar proteins and silver ions, resulting in argyrophilic staining of NOR-associated proteins.

Method (summary)
  • Hydrate section or smear with distilled water.
  • Prepare working solution: pour entire vial A into the slide container and add 10 ml of reagent B; mix gently with a clean glass rod previously rinsed in distilled water.
  • Incubate sections/slides in working solution in the dark for 30 minutes at room temperature.
  • Rinse thoroughly in three changes of high-quality distilled water.
  • Add 10 drops of reagent C to the section/slides and incubate for 1 minute.
  • Rinse in distilled water, dehydrate through ascending alcohols, clear in xylene and mount with appropriate mounting medium.


Warnings
  • Use only high-quality distilled water for all washes.
  • Do not use poly-lysine-coated slides.
  • Avoid contact with metal objects (tweezers, metal racks) during staining.
  • After mounting, store slides protected from light.
  • If reagent A is found gelled due to low-temperature storage, restore fluidity by warming the bottle under warm running water before use.
